UV Resonance Raman (UVRR) Investigations of Proteins and Peptides Secondary Structures

چکیده
Over the last 50 years numerous groups have focused on protein folding mechanisms, developing new methods to predict a protein’s native structure from its primary sequence. Nowadays, X-Ray Diffraction (XRD) and Nuclear Magnetic Resonance (NMR) are the most utilized techniques to study protein structures. However, XRD is mainly suitable for studying static state of proteins and also requires the preparation of protein crystals. On the other hand, NMR requires relatively high protein concentrations. Therefore, there is a need for other methods that give additional insight into protein structures and dynamics.

Elucidating Peptide and Protein Structure and Dynamics: UV Resonance Raman Spectroscopy.
UV resonance Raman spectroscopy (UVRR) is a powerful method that has the requisite selectivity and sensitivity to incisively monitor biomolecular structure and dynamics in solution. In this perspective, we highlight applications of UVRR for studying peptide and protein structure and the dynamics of protein and peptide folding. UVRR spectral monitors of protein secondary structure, such as the A...
متن کاملCircular dichroism and ultraviolet resonance Raman indicate little Arg-Glu side chain α-helix peptide stabilization.
Electrostatic interactions between side chains can control the conformation and folding of peptides and proteins. We used circular dichroism (CD) and ultraviolet (UV) resonance Raman spectroscopy (UVRR) to examine the impact of side chain charge on the conformations of two 21 residue mainly polyala peptides with a few Arg and Glu residues. We expected that attractions between Arg-10 and Glu-14 ...
متن کاملDirect observations of conformational distributions of intrinsically disordered p53 peptides using UV Raman and explicit solvent simulations.
We report the first experimental measurements of Ramachandran Ψ-angle distributions for intrinsically disordered peptides: the N-terminal peptide fragment of tumor suppressor p53 and its P27S mutant form. To provide atomically detailed views of the conformational distributions, we performed classical, explicit-solvent molecular dynamics simulations on the microsecond time scale. Upon binding it...
متن کاملSecondary structure polymorphism in Oxytricha nova telomeric DNA.
Tandem repeats of the telomeric DNA sequence d(T4G4) of Oxytricha nova are capable of forming unusually stable secondary structures incorporating Hoogsteen hydrogen bonding interactions. The biological significance of such DNA structures is supported by evidence of specific recognition of telomere end-binding proteins in the crystal state. To further characterize structural polymorphism of Oxyt...
متن کاملStructure and dynamics of the DNA-binding protein HU of B. stearothermophilus investigated by Raman and ultraviolet-resonance Raman spectroscopy.
The histone-like protein HU of Bacillus stearothermophilus (HUBst) is a 90-residue homodimer that binds nonspecifically to B DNA. Although the structure of the HUBst:DNA complex is not known, the proposed DNA-binding surface consists of extended arms that project from an alpha-helical platform. Here, we report Raman and ultraviolet-resonance Raman (UVRR) spectra diagnostic of subunit secondary ...
